In this study, we evaluated the in vitro and in vivo immunomodulatory properties of naturally-occurring peptides MTRP-1. This peptide significantly inhibited LPS-induced nitric oxide (NO) production in RAW 264.7 cells. It also dose-dependently reduced mRNA expression of pro-inflammatory genes such as inducible NO synthase (iNOS), tumour necrosis factor (TNF)-α, and cyclooxygenase (COX-2). In addition, immunoblotting analysis shows that nuclear levels of transcription factors such as p65, p50, c-Fos, and c-Jun were reduced by MTRP-1 treatment. Moreover, MTRP-1 inhibited the activation of both NF-κB-regulatory upstream signalling enzymes including IκBα, AKT, p85, Syk, and Src, and AP-1-regulatory enzymes including TRAF6, TAK1, MKK4/7, MKK 3/6, and IRAK1/4. Finally, MTRP-1 clearly ameliorated the inflammatory symptoms in EtOH/HCl-induced gastritis and DSS-induced colitis mouse models. These data strongly suggest that MTRP-1 can be applied as a potential natural anti-inflammatory agent with Src/Syk/NF-κB and IRAK1/4/AP-1 inhibitory properties.
